RESUMO

Ginseng is a perennial herb of the genus Panax in the family Araliaceae as one of the most important traditional medicine. Genomic studies of ginseng assist in the systematic discovery of genes related to bioactive ginsenosides biosynthesis and resistance to stress, which are of great significance in the conservation of genetic resources and variety improvement. The transcriptome reflects the difference and consistency of gene expression, and transcriptomics studies of ginseng assist in screening ginseng differentially expressed genes to further explore the powerful gene source of ginseng. Protein is the ultimate bearer of ginseng life activities, and proteomic studies of ginseng assist in exploring the biosynthesis and regulation of secondary metabolites like ginsenosides and the molecular mechanism of ginseng adversity adaptation at the overall level. In this review, we summarize the current status of ginseng research in genomics, transcriptomics and proteomics, respectively. We also discuss and look forward to the development of ginseng genome allele mapping, ginseng spatiotemporal, single-cell transcriptome, as well as ginseng post-translational modification proteome. We hope that this review will contribute to the in-depth study of ginseng and provide a reference for future analysis of ginseng from a systems biology perspective.

Circularly polarized light emission (CPLE) can be potentially applied to three-dimensional displays, information storage, and biometry. However, these applications are practically limited by a low purity of circular polarization, i.e., the small optical dissymmetry factor gCPLE. Herein, glancing angle deposition (GLAD) is performed to produce inorganic nanohelices (NHs) to generate CPLE with large gCPLE values. CdSe NHs emit red CPLE with gCPLE = 0.15 at a helical pitch (P) ≈ 570 nm, having a 40-fold amplification of gCPLE compared to that at P ≈ 160 nm. Ceria NHs emit ultraviolet-blue CPLE with gCPLE ≈ 0.06 at P ≈ 830 nm, with a 103-fold amplification compared to that at P ≈ 110 nm. Both the photoluminescence and scattering among the close-packed NHs complicatedly account for the large gCPLE values, as revealed by the numerical simulations. The GLAD-based NH-fabrication platform is devised to generate CPLE with engineerable color and large gCPLE = 10-2-10-1, shedding light on the commercialization of CPLE devices.

BACKGROUND: The verisimilitude of the reconstructed auricle and its long-term stability largely depends on the framework sculpting. This study described three kinds of minor modifications based on Firmin's way of sculpting the auricle framework and reported the clinical outcomes achieved with them. METHODS: We conducted a retrospective study of congenital microtia patients undergoing detail-improved auricular reconstruction from June 2016 to June 2020. The three kinds of minor modifications included: (1) fabricating the base frame using the 7th costal cartilage, (2) fabricating the helix and the antihelix complex using the 8th costal cartilage, and (3) fabricating the helix using the combination of the 8th and 9th costal cartilage. RESULTS: Ninety-eight patients (aged 9-27 years, 62.2% male) were included. Ninety-five patients (97.0%) adopted minor modifications 1, 2, and 3 patients (3.0%) adopted minor modifications 1, 2, and 3. All patients achieved an excellent auricle appearance and a well-laid foundation for subsequent operations. During the follow-up period, 89 patients (90.8%) were satisfied with the reconstructed auricles, 6 (6.1%) complained of hypertrophic scars in the retroauricular sulcus or pigmentation in the skin graft area, and 3 (3.1%) developed surgery-related complications. CONCLUSIONS: Three minor modifications of the auricle framework sculpting can make more satisfactory use of cartilage and adjust with the flexibility of the reconstructed auricle in different situations, making it similar to the contralateral auricle, thus, improving patients' satisfaction.

Auriculocondylar syndrome (ARCND) is a genetic and rare craniofacial condition caused by abnormal development of the first and second pharyngeal arches during the embryonic stage and is characterized by peculiar auricular malformations (question mark ears), mandibular condyle hypoplasia, micrognathia and other less-frequent features. GNAI3, PLCB4 and EDN1 have been identified as pathogenic genes in this syndrome so far, all of which are implicated in the EDN1-EDNRA signal pathway. Therefore, ARCND is genetically classified as ARCND1, ARCND2 and ARCND3 based on the mutations in GNAI3, PLCB4 and EDN1, respectively. ARCND is inherited in an autosomal dominant or recessive mode with significant intra- and interfamilial phenotypic variation and incomplete penetrance, rendering its diagnosis difficult and therapies individualized. To raise clinicians' awareness of the rare syndrome, we focused on the currently known pathogenesis, pathogenic genes, clinical manifestations and surgical therapies in this review.

BACKGROUND: For patients with a congenitally defective type of earlobe cleft, repair techniques similar to those utilized for earlobe lacerations are not suitable due to the presence of certain tissue defects. However, traditional earlobe reconstruction techniques imply the need to form complex flaps with adjacent normal tissues, which may lead to many complications. OBJECTIVES: In this study, we developed a technique to repair the earlobe with residual lobular tissue based on the characteristics of congenitally defective earlobe clefts. METHODS: We designed a triangular random flap out of the remnant lobular tissue, rotated and embedded into the edge of the remnant earlobe to repair the earlobe. RESULTS: In the past 3 years, this technique was applied to 15 patients with congenitally defective earlobe clefts. With this technique, the new earlobe achieved the desired aesthetic results, such as smooth edges and fullness of form, without complicated and unnecessary incisions. It also solved the aesthetic problem of residual lobular tissue on the face. CONCLUSIONS: Based on the characteristics of the congenitally defective type of earlobe cleft, we innovatively proposed the utilization of residual lobular tissues for earlobe repair, and the feasibility and multiple advantages of this technique have been demonstrated in practice.

OBJECTIVE: The aim of this meta-analysis was to evaluate the effect of whole-body vibration training on lower limb motor function in children with cerebral palsy in randomized-controlled trials (RCTs). METHODS: Two independent reviewers systematically searched the records of nine databases (PubMed, Cochrane, Web of Science, EMBASE, CNKI, etc.) from inception to December 2022. Tools from the Cochrane Collaboration were used to assess risk of bias. Standard meta-analyses were performed using Stata 16.0 and Revman 5.3. For continuous variables, the arms difference was calculated as the weighted mean difference (WMD) between the values before and after the intervention and its 95% confidence interval (95% CI). RESULTS: Of the 472 studies identified, 13 (total sample size 451 participants) met the inclusion criteria. Meta-analysis showed that WBV training could effectively improve GMFM88-D [WMD = 2.46, 95% CI (1.26, 3.67), P<0.01] and GMFM88-E [WMD = 3.44, 95% CI (1.21, 5.68), P = 0.003], TUG [WMD = -3.17, 95% CI (-5.11, -1.24), P = 0.001], BBS [WMD = 4.00,95% CI (3.29, 4.71), P<0. 01] and the range of motion of ankle joint and the angle of ankle joint during muscle reaction in children with cerebral palsy. The effect of WBV training on 6MWT walking speed [WMD = 47.64, 95% CI (-25.57, 120.85), p = 0.20] in children with cerebral palsy was not significantly improved. CONCLUSION: WBV training is more effective than other types of conventional physical therapy in improving the lower limb motor function of children with cerebral palsy. The results of this meta-analysis strengthen the evidence of previous individual studies, which can be applied to the clinical practice and decision-making of WBV training and rehabilitation in children with cerebral palsy.

Avoiding excessive agricultural nitrogen (N) use without compromising yields has long been a priority for both research and government policy in China1,2. Although numerous rice-related strategies have been proposed3-5, few studies have assessed their impacts on national food self-sufficiency and environmental sustainability and fewer still have considered economic risks faced by millions of smallholders. Here we established an optimal N rate strategy based on maximizing either economic (ON) or ecological (EON) performance using new subregion-specific models. Using an extensive on-farm dataset, we then assessed the risk of yield losses among smallholder farmers and the challenges of implementing the optimal N rate strategy. We find that meeting national rice production targets in 2030 is possible while concurrently reducing nationwide N consumption by 10% (6-16%) and 27% (22-32%), mitigating reactive N (Nr) losses by 7% (3-13%) and 24% (19-28%) and increasing N-use efficiency by 30% (3-57%) and 36% (8-64%) for ON and EON, respectively. This study identifies and targets subregions with disproportionate environmental impacts and proposes N rate strategies to limit national Nr pollution below proposed environmental thresholds, without compromising soil N stocks or economic benefits for smallholders. Thereafter, the preferable N strategy is allocated to each region based on the trade-off between economic risk and environmental benefit. To facilitate the adoption of the annually revised subregional N rate strategy, several recommendations were provided, including a monitoring network, fertilization quotas and smallholder subsidies.

Paddy fields in China contributed to one third of the global cropland ammonia (NH3) emission inventory, while rice accounted for half of cereal consumption, necessitating exhaustive considerations of the balance between NH3 emissions abatement and food demand. The concept of yield-scaled emission intensity (emissions per unit crop production) has the potential to guide sustainable intensification strategies, yet its application to NH3 emissions remains poorly understood. Here, by constructing novel crop-specific models for single rice production and NH3 emissions in the Middle and Lower Yangtze River Basin (LYRB) as a case study, the relationships between fertilizer N application and yield-scaled NH3 were estimated. Contrary to our hypothesis of a tipping point, our results showed that yield-scaled NH3 curves could not directly identify optimal nitrogen (N) rates. However, the benefit of lower N fertilizer rate on NH3 abatement consistently outweighed the risk of yield loss. The exponential relationships between yield-scaled NH3 and N surplus allowed us to estimate the N surplus criterion as 15.6 kg N ha-1 (or 190 kg N ha-1 fertilizer N rate) for the LYRB. Under the N surplus criterion, NH3 emissions can be reduced by 23-27% without severely impacting rice yield, compared to the N rate required for the highest yield. Moreover, five major controlling factors for yield-scaled NH3 were estimated by random forest models, ranked in order of importance as N rate, total N, K rate, mean annual precipitation, and soil organic carbon. Among the agricultural practices (irrigation, tillage, and fertilizer management), deep placement was the most effective measure to reduce yield-scaled NH3, showing 48% reduction potential, followed by proper N splitting frequency (43%). Overall, this study highlights the efficacy of N application optimization and targeted farm management in mitigating NH3 emission while maintaining crop productivity.

Acceleration of singlet-triplet intersystem crossings (ISC) is instrumental in bolstering triplet exciton harvesting of multi-resonance thermally activated delayed fluorescent (MR-TADF) emitters. This work describes a simple gold(I) coordination strategy to enhance the spin-orbit coupling of green and blue BN(O)-based MR-TADF emitters, which results in a notable increase in rate constants of the spectroscopically observed ISC process to 3×109  s-1 with nearly unitary ISC quantum yields. Accordingly, the resultant thermally-stable AuI emitters attained large values of delayed fluorescence radiative rate constant up to 1.3×105 /1.7×105  s-1 in THF/PMMA film while preserving narrowband emissions (FWHM=30-37 nm) and high emission quantum yields (ca. 0.9). The vapor-deposited ultrapure-green OLEDs fabricated with these AuI emitters delivered high luminance of up to 2.53×105  cd m-2 as well as external quantum efficiencies of up to 30.3 % with roll-offs as low as 0.8 % and long device lifetimes (LT60 ) of 1210 h at 1000 cd m-2 .

BACKGROUND: In this article, we will introduce a modified surgical approach to correct cryptotia (by suspending the auricular cartilage to the temporal region), which allows for a more visible and consistent surgical benefit than previous techniques that simply used flap accumulation. METHODS: We use several stitches to suspend the released auricular cartilage from the root of the auricular cartilage (approximately deep in the cavum conchae and the cymba conchae cartilage) to the temporal bone periosteum in a higher position, presumably similar to the principle of leverage, thus obtaining a larger auriculocephalic sulcus and a higher superior auricular pole. RESULTS: Over the past 5 years, this new technique was applied to the auricles of 20 patients between the ages of 15 and 40 years. They all obtained a satisfactory height of the upper pole of the auricle, the auriculocephalic sulcus, and the amount of skin tissue behind the auricle. CONCLUSIONS: In response to the failure of previous techniques to provide a satisfactory auriculocephalic sulcus and superior auricular pole height, this modified technique provides more consistent support directly to the auricle by means of suspension, allowing for a deeper head sulcus and higher superior auricular pole. The main features of this technique are the convenience and stability of the results, and the ability to adjust the sulcus and the height of the auricle as needed during the surgery.

Achieving the early diagnosis of breast cancer, through ultrasensitive detection of tumor marker miRNA-155, is a significant challenge. Therefore, an ultrasensitive hairpin electrochemical biosensor based on graphite-like phase carbon nitride composite was proposed. In this paper, poly(D-glucosamine) (PDG) was used as a stabilizer and reducing agent to prepare gold nanoparticles at room temperature, and then a graphite-like phase with a two-dimensional lamellar structure carbon nitride was further combined with it to obtain the poly(D-glucosamine)/gold nanoparticles/graphite-like phase carbon nitride nanocomposite (PDG/AuNPs/g-C3N4), in order to achieve the goal of signal amplification. The specific hairpin capture probe (HP) that recognized and bound miRNA-155 was then grafted. The hairpin biosensor showed a linear range of 0.1 fM-1 pM with a detection limit of 0.05 fM using differential pulse voltammetry (DPV) electrochemical analysis. Furthermore, the excellent performance hairpin electrochemical biosensor had been applied to the detection of miRNA-155 in human serum samples with good recovery.

The current power distribution Internet of Things (PDIoT) lacks security protection terminals and techniques. Network security has a large exposure surface that can be attacked from multiple paths. In addition, there are many network security vulnerabilities and weak security protection capabilities of power distribution Internet of Things terminals. Therefore, it is crucial to conduct a scientific assessment of the security of PDIoT. However, traditional security assessment methods are relatively subjective and ambiguous. To address the problems, we propose to use the entropy-weight method and cloud model theory to assess the security risk of the PDIoT. We first analyze the factors of security risks in PDIoT systems and establish a three-layer PDIoT security evaluation index system, including a perception layer, network layer, and application layer. The index system has three first-level indicators and sixteen second-level indicators. Then, the entropy-weight method is used to optimize the weight of each index. Additionally, the cloud model theory is employed to calculate the affiliation degree and eigenvalue of each evaluation index. Based on a comprehensive analysis of all evaluation indexes, we can achieve the security level of PDIoT. Taking the PDIoT of Meizhou Power Supply Bureau of Guangdong Power Grid as an example for empirical testing, the experimental results show that the evaluation results are consistent with the actual situation, which proves that the proposed method is effective and feasible.

Soil acidity is one of the vital factors that influence organic matter transformation and accumulation. Long-term studies on the mechanisms of biochar's effects on soil organic matter (SOM) accumulation dependent on pH values are lacking. A four-year column experiment was conducted without and with biochar application (11.3 Mg ha-1 crop-1) in acid (pH = 5.24) and alkaline (pH = 8.22) soils under paddy rice/wheat annual rotation. To explore organic matter accumulation mechanisms, SOM pools were extracted (physical-chemical fractionation) and their chemical structures were analyzed using advanced solid-state 13C nuclear magnetic resonance (13C NMR) techniques. Biochar increased the proportion of aromatic carbon (C) in all SOM pools, which led to an increased C content in two soils. The elevated pH after biochar application (∆pH = 1.03) increased Fe (III) oxidation and precipitation, and therefore, stimulated amorphous Fe content in 53-µm pool in the acid soil. This change increased the interaction between organic compounds and Fe (hydr)oxide, which impeded bacteria access to substrates, and in turn, promoted SOM accumulation in the acid soil. Conversely, low Fe (hydr)oxide availability resulted in the decomposition of the labile substrates (di-O-alkyl C, NCH, and OCH) in mobile humic acids via microbial respiration, thereby lowering the effect of SOM sequestration in the alkaline soil. Our study revealed that organic matter accumulation after biochar amendment is not solely dependent on the chemical recalcitrance of biochar, but also is controlled by the transformation of Fe (hydr)oxide in SOM pools.

Application of biochar (BC) derived from rice straw has generated increasing interest in long-term storage of soil organic carbon (SOC), however its carbon (C) sequestration potential vary widely among agricultural soils despite the same BC dose used. These discrepancies in the ability of soils to sequester C after BC application are poorly understood. Metabolic quotient (qCO2) is a reflection of "microbial efficiency" and linked to SOC turnover across ecosystems. Therefore, we investigated the SOC sequestration and qCO2 in a Yellow River alluvium paddy soil (YP) and a quaternary red clay paddy soil (QP) under rice-wheat annual rotation following 4-year of BC application rate of 11.3 Mg ha-1 per cropping season. BC application consistently brought 65.3 Mg C ha-1 into the soils over 4-year experimental period but increased SOC by 57.6 Mg C ha-1 in YP and 64.5 Mg C ha-1 in QP. Calculating SOC mass balance showed 11.7% of BC-C losses from YP and only 1.16% from QP. BC application stimulated the G+ bacterial, fungi, and actinomycetes by increasing O-alkyl C content in YP, while decreased the same microorganisms by decreasing anomeric C-H content in QP. Importantly, higher clay and amorphous Fe (Feo) contents in QP after BC application protected SOC from further decomposition, which in turn decreased microorganisms and resulted in higher SOC sequestration than YP. Our results indicated that soil properties controlled the extent of SOC sequestration after BC application and site-specific soil properties must be carefully considered to maximize long-term SOC sequestration after BC application.

Converting straw to biochar (BC) followed by successive application to soil has been increasingly suggested as a multi-win approach for soil fertility improvement, carbon (C) sequestration and efficient disposal of straw residues in intensive cropping agroecosystems. However, different soil types response differently in terms of crop growth and non-CO2 greenhouse gas (GHG) emissions after BC application. Furthermore, few studies have comprehensively evaluated the net global warming potential (GWP) and net ecosystem economic benefits (NEEB) after long-term BC incorporation across representative soil types in China. A five-year outdoor column experiment was conducted using three rice-wheat rotated paddy soils and three millet-wheat rotated upland soils developed from different parent materials. Rice straw BC application rates of 0, 2.25 and 11.3 Mg ha-1 were used in each crop season with identical doses of NPK fertilizers. Compared with the no BC controls, BC significantly boosted crop growth, enhanced C sequestration, and decreased cumulative N2O and CH4 emissions in all six soils over five rotation cycles. The response of the upland soils to BC was better in terms of crop growth and N2O mitigation, whereas the soil organic carbon (SOC) increment and CH4 mitigation were less effective compared with the paddy soils. Net GWP decreased 0.6-19 fold after BC application; however, given the low trade price of CO2 (0.21 × 103 CNY Mg-1), only a small contribution was made in terms of C costs to the NEEB. The BC-induced NEEB was mainly dependent on grain yield gains and BC costs. These findings highlight that widespread adoption of successive straw BC application to farmland requires an increase in crop yield and substantial lowering of the BC cost regardless of the soil type. From the standpoints of agronomics, environment and economics, acid upland soil shows most potential in terms of BC application.

OBJECTIVE: To assess the sensitisation effects of DDR inhibitors combined with conventional chemotherapeutics agents (cisplatin et al) in a drug-resistant ovarian cancer cell line(OVCAR-8). METHODS: Inhibitors of DDR regulators with cisplatin were applied to challenge OVCAR-8, and evaluated the DNA damage response (DDR) and cytotoxic effects of different combination of chemicals. Inhibition of proliferation to OVCAR-8 of different drugs was evaluated by MTT assay. The activation of phosphorylation of histone family 2A variant (yH2AX) and p53 binding protein 1 (53BP1) in OVCAR-8 were evaluated by immunofluorescence to observe their ability of recruitment and forming foci at DNA damage site. RESULTS: We observed that combined treatment of ataxia-telangiectasia mutated (ATM)/ATM and Rad 3-related (ATR) inhibitor and cisplatin can suppress the activation of damage repair mechanisms and weakened the proliferative activity of OVCAR-8 cells (P<0. 01) ; ATR pathway was suppressed and the signal of γH2AX weakened and cell survival rate significantly reduced when combination therapy of HU and Wortmannin (P < 0.05); poly ADP-ribose polymerase (PARP) inhibitor could not enhance chemosensitivity in OVCAR-8 cells when combined with cisplatin (P > 0.05). CONCLUSION: We substantiated that appropriate inhibitors of DNA damage response may have a potential to improve the anti-tumor effect of conventional chemotherapy drugs and prevent drug resistances.
